What Is The Risk Of Deafness In Case Of A Consanguineous Marriage?

my parents are not cousins, or related in anoy other way nor do they have any physical disorders that we know about. when my big brother was born he was deaf, the same with the second brother. When i was born (female) i was completley normal. then finally my little brother was bron and he also was deaf. so all 3 males in the fmaily were deaf and the only girl (me) is not. now my brother is on holiday in Pakistan and he has decided he wants to marry my mum's sister's (who has passed away) daughter, therefore my brothers cousin.



The only thing that is keeping my dad from agreeing is because he is deaf and is getting married to a first cousin. Would you be able to explain the egenral risks involved if they were to get married and have kids in the future. When my brother comes back from Pakistan we will be snding him for genetics testing as well as getting a sample of the girls blood so that both set of genetics can be put together to see any risks etc. but for just now i need an answer of a general nature.

Thank you for following up. May be you know closely marriage increases risk of genetic disease. It is clearly seen that your brother has X-linked deafness, because it is express only at boys. Chance that his future wife will be carrier of pathogenic X-chromosome -50%. Their risk for kids will compose:
-25% girls with normal hearing, but carriers of pathogenic gene
-25% completely deaf girls
-25% absolutely normal boys
-25% absolutely deaf boys
The genetics strongly disagree with consanguineous marriage.
