Can Missing Duphaston Once Cause Spotting?

Hi Doc, I had my last periods on Sep 21st. Trying for pregnancy.My follicle has ruptured on this Oct 6th and my Doc prescribed me one tab per day of Duphaston before bed for 13days.We intercoursed everyday. And I missed one dose on Oct 12th night and as I woke up next day I recollected and took the tablet in the morning and again I didnt take it before bed on the same day.So from the next day i continued taking it in the morning sessions. And i saw tiny brown spot on Oct 12th night. And little more spots on Oct 13th and 14th and it stopped by Oct 15th with tiny spots. Now out of my 13 tablets of duphaston I missed one so what should i do? Should i take that leftover tablet or not? And is this the reason behind my brown spots or am I Pregnant . So when can i expect my periods this month ?
