Brief Answer:
Can be due to infection or injury in the area.

Detailed Answer:
Hello,
Thanks for posting your query.
The severe pain at the back of thigh or behind the knee can be due to an abscess formation or a trauma in the area. This needs a direct examination and investigations like MRI scan to detect the soft tissue abnormalities in the area.
If the pain is gradually increasing then it is best to consult your physician for a direct examination and proper investigations.
I hope this answers your query.

Pain is most severe when I first try to stand on the leg and then the pain goes down after I am up and moving. The muscle that is hurting right now i shalf way up the thigh in the back and the pain radiates from side to side. The second pain is as I said behind the knee and in the top of the muscle going down the leg. I have started to now get XXXXXXX horses in the main calf muscle. I take 400 mg MG daily due to low magnesium levels.

Icing and rest will not help you are saying

Brief Answer:
Electrolyte imbalance likely.

Detailed Answer:
Hello.
Thanks for writing again. XXXXXXX horses are due to muscle spasms and are directly related to the electrolyte imbalance. You need to get your electrolytes evaluated again and see if any more supplements are needed.
The pain behind knee can be related to a knee pathology or osteoarthritic changes. These can be best detected on an MRI scan so that specific treatment can be started.
Hope my answer is helpful.
