I have gone through your query and I can understand your concerns.

First of all PSA(prostate specific antigen) is prostate specific, not cancer specific. PSA, is a protein released from inflamed prostate. Manipulations like colonoscopy and sometimes enema can cause release of PSA from the prostate and cause of false positive results.
Enema cause very slight insignificant increase compared to other procedures like colonoscopy and sigmoidoscopy.

PSA is considered as a screening test only. We would not make a diagnosis of prostate cancer solely based on PSA. If we detect a high PSA value,we would go for a biopsy to confirm the diagnosis.

A PSA level of 6 is considered high for your age.
For men in their 50s or younger, a PSA level should be below 2.5 .

As I said PSA is not specific for cancer. We have to rule out conditions like BPH(Benign prostatic hypertrophy) ,Prostate infection (prostatitis) ,Urinary tract infection.
You can re do the PSA test after abstaining from sex for few days as sexual intercourse can cause falsely high PSA level.
I suggest you to consult a urologist and get a biopsy done to confirm a diagnosis.
