Question: i have two clear pus filled blisters on penis right below mushroom. allergic to penecillon and iodine

Can you upload a good quality picture of the area?

If the blisters are absolutely clear, this could be either from a herpetic infection (such as HSV II or I) or it could be from an irritation. Usually herpetic infections are quite painful. There are antiviral medications to decrease the severity and duration of HSV (although no cure).

However if there is visible pus, this is more likely to be a bacterial infection. For a local, small bacterial infection you can sometimes treat it with hot soaks, hot compresses (not so hot it burns your skin though). However, if it spreads and gets bigger, then you need to go in to see a doctor in person.

the pus was visible its gone away and it wasnt painful just irritating and i had madturbated a lot a few nights before so i do think its feom irritation

Yes, this can happen.

Skin friction can sometime cause an irritation and body reacts with blisters (such as you can get on the bottom of your foot from new shoes). You may want to take a break for a day or two until it completely heals over, and be sure to use a good lubricant in future, such as AstroGlide or KY gelly or other personal lubricants. These can decrease trauma from friction.

If the blisters were not very painful, the chances of them being herpetic are low. So yes, the likelihood that it was from irritation is higher. Without seeing them when they were most active, it is hard to know. Are you at risk for STDs? (ie unprotected sexual encounters in past several weeks?)
