Tuberculosis ( if active or took treatment , report), If first iui failed try for 2nd one.
As far as concerned about PCOS , if successful ovulation is there , pregnancy will be there.
Estrogen, FSH/LH ratio , androgen levels play an important role in pregnancy , so blood report of these hormones upload.

The spotting would not in any way signify a pregnancy.
IT could be a scantier than normal period.
The best and most reliable way to diagnose a pregnancy is to take a pregnancy test, if it has already been 2 weeks since your IUI.
Also a blood B HCG test would also let you know reliably if there is a pregnancy.

Hi ! Beta HCG levels vary differently in different coditions. It is difficult to say what is normal for each individual. I would suggest you to go for an antenatal ultrasound of abdomen after consulting your gynaecologist to logically correlate the level of HCG with the duration or pregnancy. Take care.
