Hello,
If she has always been regular with her cycles and if repeated tests for pregnancy done with proper precautions are negative, it cannot be pregnancy. Yet, a clinical examination by a specialist, an estimation of serum beta-hCG titres and a trans-vaginal sonogram are necessary to come to a conclusion. If pregnancy is excluded, hormonal imbalances need to be suspected and managed accordingly. A home pregnancy test needs to be performed 3-7 days after the missed period with a fresh early morning sample of urine. Hope this helps.
