Me and my girlfriend had sex 10 days ago. She is supposed to have just one antibiotic right after sex to prevent uti. We used a condom but for the first five minutes we didn't. She is 4 days late with her period.

tHE CONCERN REGARDING HER PREGNANCY IS WELL UNDERSTOOD. As she is 4 days late for her menses the best way to rule out pregnancy at present would be to undertake a serum beta hcg test. This test if negative or hcg levels less than 1ng/dl shall rule out pregnancy completely.
As you were not protected initially for 5 min, the chances that pre cum also may cause fertilisation and pregnancy and as she was in her fertile period, there could be a possibility.
So the best way to rule out pregnancy as of now is serum beta hcg levels.

Increased mucoid discharge is not exactly the sign of impending menses, rather the premenstrual symptoms are usually in form of mild pain in lower abdomen, nausea, headache, mood swings, increased frequency of urine abdominal cramps.
If you want you may wait for another 4 days and then you can do the urine pregnancy test if you desire and till then there are chances that the menses may start as well, so you may either wait or if anxious then you can get the serum beta hcg test for results.

Can just one antibiotic pill delay menses that much?

No it is unlikely that a single dose shall delay the menses, but the concern is not antibiotic but the reason or illness why she was given an antibiotic. ILLNESS during menstrual cycle can cause stress in women and stress can easily delay her menses by a week or so.
So you may wait if you desire.
